NCL, Celebrity, Princess and Cunard do South America cruises. I am sailing from Santiago/Valparaiso to Buenos Aires via Cape Horn...NCL Crown does this route and reverse all winter, Celebrity Infinity does it too, Regal Princess runs from Miami down to Rio and on to Valparaiso and Cunard QM2 will be starting some SA itineraries aswell.
If you take a quick look at the 'Norwegian Cruise Lines Roll Call' section here, I have typed out my entire itinerary.
Celebrity also do the Galapagos Islands with their ship Xpedition, other companies go there too.
There are alot of repositioning cruises that leave from Miami or Philly and travel through the Panama Canal and on down the west coast of SA and then around the Cape and back up the east coast. P&O ship Minerva and a couple others such as Raddisson sail up to Manaus on the Amazon too.
The easiest way to find out what is what is to do a search online using 'cruises South America' as keywords.
Only my opinion, but I think in the next few years, we'll see SA becoming a more frequented destination for cruise ships.
I would think that plenty of folks would like to visit there, but would not consider just flying in because of the political climate and reports of kidnappings in some of the contries there. However, a cruise ship provides what might be seen as a 'safer' way to see at least snippets of that big continent to our south.
Hanna, we are getting ready to book our own SA cruise too.
There seem to be about 6 standard itineraries on the bigger cruise lines.
From Florida or the Caribbean thru the Panama Canal, down the west coast of SA down to Valparaiso Chile.
And the reverse
From Valparaiso all the way south, go around Cape Horn and then up to Buenos Aires, Argentina.
And the reverse
During Carnival time, Round trip from Buenos Aires to Rio de Janeiro, Brazil for the festivities.
From Buenos Aires north to the Caribbean or Florida.
And the reverse.
From Florida or the Caribbean down to Brazil, sail down the Amazon River to Manaus, Brazil.
And the reverse.
Oh well, that's 5 and 9 if you count the reverses. Some of the luxury lines may have additional itineraries too.
We picked Buenos Aires to Valparaiso, because my DH wants to see Cape Horn, and because we can book a Celebrity cruise on our next cruise.

...and NCL and Cunard and Orient Lines too...
Orient's Marco Polo goes to Antartica too....along with Princess.
Thank you Ally for keeping me straight!!! How could I forget NCL? I think I was counting Cunard as one of the luxury lines, but I have to admit, I'm not familiar with Orient.
Always good to have more options!

The Marco Polo was built in Russia in 1965, she has a fully strengthened hull for ice breaking, her original name was Alexandr Pushkin. She is beautiful, classic liner. Orient is part of Star, and in the NCL family but she trades alone.
I was just reading in a magazine last night of the increasing popularity of South American cruises. As well as an increasing popularity of South American, including a jaunt down to Antartica cruise.
